$IONS down almost 17% this morning on failed trial news in partnership with Astrazenca. The data is below. You decide. What was actually lost vs. what's intact: Lost: The ATTR-CM (cardiomyopathy) indication, the big half of the ~$35B ATTR market. But three things matter here: - It was future optionality, not current revenue, ATTR-CM contributed $0 to today's P&L. Literally zero current revenue was destroyed this morning. - Ionis's economics there were a royalty, not full ownership. - The competitive bar was already brutal Alnylam's Amvuttra got ATTR-CM approval with a CV-outcomes win, plus BridgeBio (acoramidis) and Pfizer (tafamidis) entrenched. Eplontersen was a late 4th entrant hoping to differentiate. The Streetwas not crediting this as a sure thing. On fundamentals, this is an overreaction: ~$2.5B of market cap (18% of a ~$13.9B company) was erased over an asset that generated $0 current revenue, was a royalty-only long shot against entrenched competition, and doesn't touch any of the drivers that actually carry the stock. We want to bring your attention to IONS. IONS is a buyout candidate. Today's FDA approval just removed the last major binary risk. Here is the thesis. Ionis Pharmaceuticals is the pioneer of RNA-targeted medicine. Thirty years of antisense technology development. Six marketed medicines. Two more independent launches in 2026. Four partner launches by end of 2027. AstraZeneca, Biogen, Roche, and Otsuka are all paying to access the platform. Today the FDA approved TRYNGOLZA (olezarsen) for severe hypertriglyceridemia. Priority Review. Breakthrough Therapy designation. The first therapy approved for this indication. 1 million high-risk patients initially, 3 million as primary care expands. Management has already guided TRYNGOLZA peak net sales above $2 billion for this indication alone. The drug generated $105 million in 2025 from a much smaller FCS patient population. Today's approval opens the much larger sHTG market. The CARDIO-TTRansform Phase 3 data with AstraZeneca is coming H2 2026. More than 1,400 patients. The largest ATTR-CM study ever conducted. If it reads positive, Ionis becomes significantly more valuable overnight. The buyout case. AstraZeneca is already inside. Strategic collaboration on eplontersen globally. They have visibility into the pipeline, the technology, and the commercial execution. Converting a partnership to an acquisition is the path of least resistance. The former CEO of Alexion, which AstraZeneca acquired, just joined the Ionis board of directors this month. The market cap is $11 billion. For AstraZeneca, Novartis, Roche, or Pfizer that is one quarter of free cash flow. Six marketed medicines, two 2026 launches, four partner launches by 2027, a $2 billion peak sales drug approved today, and a platform that generates drugs for decades. The RNA platform is what they are buying. Not TRYNGOLZA. The platform. The options market saw it today. August call volume hit 3,075 contracts versus 439 in July. Seven times the call interest in the August window, which covers the CARDIO-TTRansform data. The put/call ratio at July expiry is 0.24, one of the most bullish readings you will see. Someone is positioned for a catalyst before August expiration. The trade. Buy IONS at $76.52. Stop at $72, below institutional support. Target $100-110 on M&A. Target $95-105 on standalone TRYNGOLZA launch over 12-18 months. $20-30 upside versus $4.50 downside. 4 to 6x reward to risk. No options decay. No expiration. The FDA approval today was the last binary risk. The platform is proven. The pipeline is loaded. The $2 billion drug just got approved.
